Single cell transcriptomics identify AHR and CD9 as markers of CD14 +atypical B cells

چکیده
Abstract B lymphocytes are adaptive immune cells responsible for the production of antigen specific antibodies and release pro- anti-inflammatory cytokines. Pro-inflammatory cells, often termed atypical (at cell) a heterogenous population associated with aging, infection, autoimmunity. Aryl hydrocarbon receptor (AHR) is ligand-activated transcription factor important xenobiotic metabolism. However, AHR heavily implicated in cell differentiation as regulator fate decisions. at believed to represent an alternative lineage but role not known. We have reported that human, primary CD5 +innate-like (ILB) preferentially sensitive modulation by AHR-activation compared −B cells. +ILB enriched IgM memory B1 marginal zone regulatory used Single transcriptomics elucidate expression human naïve, circulating CD19 +B isolated directly from PBMC protein. found correlated CD14and were myeloid gene signature. Single Cell Transcriptomics: Methods and Applications
Traditionally, gene expression measurements were performed on "bulk" samples containing populations of thousands of cells. Recent advances in genomic technology have made it possible to measure gene expression in hundreds of individual cells at a time. As a result, cellular properties that were previously masked in "bulk" measurements can now be observed directly. In this review, we will survey...

The emerging single-cell RNA-Seq (scRNA-Seq) technology holds the promise to revolutionize our understanding of diseases and associated biological processes at an unprecedented resolution. It opens the door to reveal intercellular heterogeneity and has been employed to a variety of applications, ranging from characterizing cancer cells subpopulations to elucidating tumor resistance mechanisms. ...

An understanding of the distinct families of molecules present on different cells of the immune system provides the tools for distinguishing these cell types in both diagnosis and therapy, as well as understanding the molecular basis for many lymphoid cell functions. Diagnostic MARKERS for T-cells include the T-cell receptor (TcR) with its associated CD3 signaling complex as well as CD5 and the...
متن کاملMoore's Law in Single Cell Transcriptomics
The ability to measure the transcriptomes of single cells has only been feasible for a few years, and is becoming an extremely popular assay. While many types of analysis and questions can be answered using single cell RNA-sequencing, a central focus is the ability to survey the diversity of cell types within a sample. Unbiased and reproducible cataloging of distinct cell types requires large n...
متن کاملSingle-cell transcriptomics for microbial eukaryotes
One of the greatest hindrances to a comprehensive understanding of microbial genomics, cell biology, ecology, and evolution is that most microbial life is not in culture. Solutions to this problem have mainly focused on whole-community surveys like metagenomics, but these analyses inevitably loose information and present particular challenges for eukaryotes, which are relatively rare and posses...
